Apoptosis란 외부인자의 자극이나 내적인 변화에 의해서 그 세포들이 자발적으로 죽음에 이르는 것으로 programmed cell death(PCD)라고도 말합니다. 이것은 생물체의 발생 과정 중에 효용성을 상실한 세포, 암세포, 바이러스에 감염된 세포, 자가면역 세포 등과 같이 불필요하거나 위험한 세포들의 제거 기전으로 사용되고 있습니다. 반면, necrosis는 갑작스런 세포의 죽음으로 염증을 유발하여 괴사를 일으키는 것을 말합니다. |

Apoptosis의 기능

Cell termination: 바이러스 감염, 스트레스 등의 원인에 의해 damage를 입은 세포는 스스로 제거되어 나머지 조직들을 보호하게 합니다. Homeostasis: Adult organism에서 세포수는 적절한 세포 사멸과 세포분열을 통해 유지됩니다. 여기서, Apoptosis는 세포 사멸에 관여하는 기전입니다. 세포 분열 > 세포 사멸 : Tumor Development: 식물이나 동물의 조직 발생학에 있어서 apoptosis는 아주 중요한 역할을 합니다. 가끔 조직이나 기관의 발생 단계에서 불균형 분화 또는 분리가 진행될 때, apoptosis는 이를 정상적인 형태로 되돌려 주는 역할을 합니다. |

A1: 아닙니다. Cathepsin B는 caspase-independent 과정에 대해서도 PS를 유도할 수 있습니다.
Q2: Apoptosis 반응에 대한 cell는 Annexin V에 대해 염색되지만, trypan blue나 다른 vital dye에 대해 염색이 되지 않는 이유는 무엇인가요?
A2: Early stage의 apoptosis는 intact membrane을 갖고 있고, trypan blue 등과 같은 dye는 membrane을 통과하지 못하기 때문에 염색이 되지 않습니다.
Q3: Sample buffer에 EDTA를 포함하고 있을 때, positive staining 결과를 얻을 수 없는 이유는 무엇인가요?
